Usually they just charge a deposit (which is equal to the first night) and the balance when you check out. If booked directly with the hotel you can usually cancel within 24 or 48 hrs prior to arrival with no charge (they refund your credit card) Check the fine print if booking online or ask the reservation desk.

My experiences is that when you book, they charge you the first night, you have anywhere from 48-72 hrs depending on which hotel to cancel and get a full refund. When you check out at the end of your stay, they charge you the remainder of your charges.
If you book on a site like expedia, or hotels.ca or priceline I am not sure you can cancel or change without charges, check the fine print if you are not booking direct with the hotel.

I have booked hotels through Expedia and travelocity. They charge the full amount immediately and it is non-refundable.
Last summer I booked all of our Vegas hotels through the hotels themselves, was charged one night for each reservation, and the remainder owing at checkout time.
Last month I made our hotel and flight reservations through MGMmiragevacations. They deducted the full amount upfront. I did have the option to purchase travel insurance which would cover a refund on the hotel room but not the flight.
